بررسی پلی‌مورفیسم R577X در ژن ACTN3 درکشتی‌گیران

چکیده
مقدمه و هدف: عملکرد ورزشی یک فنوتیپ پیچیده است که توسط چندین عامل محیطی از جمله رژی م­غذایی، ورزش و عوامل اجتماعی تعیین می‌ گردد. عوامل ژنتیکی نیز ممکن است به تفاوت بین فردی در عملکرد ورزشی افراد کمک کند. هدف از پژوهش حاضر بررسی فراوانی آلل‌های پلی‌مورفیسم ژن ACTN3 در بین کشتی‌ گیران مبتدی نوجوان اردبیلی بود.
مواد و روش­ ها: آزمودنی‌‌های این تحقیق 15 کشتی‌گیر مبتدی نوجوان پسر (با سن 1.53±13.07 سال و وزن 7.37± 48.66 کیلوگرم) از شهر اردبیل بودند. روش اندازه‌گیری پلی‌مورفیسم ژن ACTN3 با استفاده از واکنش زنجیره‌ای پلیمراز و توالی‌یابی مستقیم DNA از طریق نمونه بزاقی تعیین و مورد مقایسه قرار گرفت. داده‌ها با آزمون‌ کای‌دو برای نیکویی برازش و در سطح معنی‌داری 0.05>P با استفاده از نرم افزار SPSS تحلیل شدند.
یافته‌ها: نتایج نشان‌ داد که تفاوت معنی‌داری در فراوانی‌های پلی‌مورفیسم ژن ACTN3 وجود داشت (0.001>P). فراوانی نسبی ژنوتیپ ‌های RR و RX به­ ترتیب برابر 80% (تعداد=12) و 20% (تعداد=3) بود. همچنین، فراوانی نسبی آلل‌های R و X به ترتیب برابر 90% (تعداد=27) و 10%
(تعداد=3) بود.
بحث و نتیجه‌گیری: به طور کلی، نتایج نشان‌دهنده‌ برتری آلل R در پلی‌مورفیسم ژن ACTN3 در کشتی‌گیران نوجوان مبتدی پسر اردبیلی بود. به نظر می‌رسد ارزیابی فراوانی‌های پلی‌مورفیسم این ژن‌ها ممکن است دراستعدادیابی رشته­ های ورزشی از جمله کشتی در رده‌های سنی پایه، تأثیر‌گذار باشد.

چکیده English

Introduction and Purpose: Athletic performance is a complex phenotype that is determined by several environmental factors such as diet, exercise and social factors. Genetic factors may also contribute to the inter-individual differences in athletic performance. The aim of the current study is to investigate the frequencies of alleles of polymorphism of ACTN3 gene among Ardabilian adolescent beginner wrestlers.
Materials and Methods: The subjects in this research were 15 beginner teen wrestler boys (with 13.07±1.53 years old and 48.66±7.37 kg weight) from Ardabil city. The method of measuring ACTN3 gene polymorphism using polymerase chain reaction and direct DNA sequencing through saliva sample was determined and compared. The data were analyzed by goodness of fit chi-squared test at a significant level P<0.05 using SPSS 22 software.
Results: The results showed that there was a significant difference in the frequencies of ACTN3 gene polymorphism (P<0.001). The relative frequencies of RR and RX genotypes were 80% (n=12) and 20% (n=3), respectively. Also, the relative frequencies of R and X alleles were 90% (n=27) and 10% (n= 3), respectively.
Discussion and Conclusion: In general, the results showed the superiority of the R allele in ACTN3 gene polymorphism in the Ardabilian beginner teen wrestler boys. It seems that the evaluation of the frequencies of polymorphism of genes may be effective in choosing the right sports field in basic age classes.
