تأثیر 4 هفته تمرین هوازی تداومی باحجم‌های مختلف وگرسنگی بر بیان برخی ژن‌های دخیل در میتوفاژی کبد رت‌های سالم نر نژاد ویستار

چکیده
مقدمه و هدف: افزایش تعداد میتوکندری­ های معیوب و تداوم آن می ­تواند موجب آپوپتوز و نکروز سلول­ ها گردد. تمرینات ورزشی منظم به همراه محدودیت زمان مصرف کالری دریافتی (TRE) می ­تواند در بهبود آسیب و عملکرد میتوکندری نقش مهمی داشته باشد. لذا هدف این مطالعه بررسی تاثیر 4 هفته تمرین هوازی تداومی وگرسنگی بر بیان برخی از ژن­ های دخیل در میتوفاژی شامل  PINK1و BNIP3و برخی از عوامل موثر برعملکرد سلامت کبد از جمله آنزیم ­های کبدی ALT و AST و نیمرخ لیپیدی رت های سالم می ­باشد.
مواد و روش ­ها: مطالعه حاضر از نوع تجربی می باشد. 30 سر رت نر سالم نژاد ویستار 20-18 هفته‌ای با میانگین وزن بدن 25.34 ±330.40 گرم که از انیستیتو پاستور ایران خریداری شده بودند به عنوان نمونه تحقیق انتخاب و به طور تصادفی ساده در 6 گروه ‌5 تایی: کنترل، گرسنگی، 3 روز تمرین، 5 روز تمرین، گرسنگی + 3 روز تمرین، گرسنگی + 5 روز تمرین تقسیم شدند. تمرین ورزشی در مدت 4 هفته به تعداد 3 و 5 روز در هفته به مدت 60-45 دقیقه روی تردمیل انجام شد. 24 ساعت پس از آخرین جلسه تمرینی و 14 ساعت ناشتایی بافت­ برداری انجام و خون­ گیری بلافاصله از قلب صورت گرفت. روش آماری تحلیل واریانس یک راهه و آزمون تعقیبی LSD نیز برای مقایسه داده ها استفاده گردید.
یافته ها: شاخص های وزن، کلسترول و ALT در همه گروه­ها در مقایسه با گروه کنترل کاهش معناداری نشان داد (0.05>P). همچنین، دو گروه گرسنگی + 3 روز تمرین وگرسنگی + 5 روز تمرین در مقایسه با سایر  کاهش بیشتری در این شاخص ­ها ایجاد کردند، اما بین آنها تفاوت معناداری مشاهده نشد. شاخص تری­ گلیسرید در همه گروه­ ها در مقایسه با گروه کنترل کاهش معناداری یافت. بین سایر گروه ­ها تفاوت معناداری مشاهده نشد. شاخص AST در همه گروه ­ها در مقایسه با گروه کنترل کاهش معناداری داشت
(0.05>P). گروه­ های 3 و 5 روز تمرین، گرسنگی + 3 روز تمرین و گرسنگی+ 5 روز تمرین در مقایسه با سایر گروه‌ها کاهش بیشتری را نشان دادند و بین این گروه‌ها تفاوت معناداری مشاهده نشد. شاخص  PINK1در گروه ­های 5 روز تمرین، گرسنگی + 3 و 5 روز تمرین در مقایسه با گروه کنترل و گروه گرسنگی کاهش معناداری یافت (0.05>P). بین سایر گروه­ ها تفاوت معناداری مشاهد نشد. شاخص BNIP3 درگروه­ های 3 روز تمرین، 5 روز تمرین و گروه گرسنگی + 3 روز تمرین در مقایسه با گروه کنترل افزایش معنی­ داری نشان داد (0.05>P). گروه 5 روز تمرین افزایش بیشتری را در مقایسه با گروه گرسنگی + 3 روز تمرین ایجاد کرد. بین گروه­ های گرسنگی و گرسنگی + 5 روز تمرین در مقایسه با گروه کنترل تفاوت معناداری مشاهده نشد.
نتیجه گیری: چهار هفته تمرین هوازی تداومی و گرسنگی توام با ورزش، با تنظیم کاهشی وضعیت نیمرخ لیپیدی و افزایش بیان ژن BNIP3 می­ تواند اثرات مثبتی بر سلامت کبد با وضعیت میتوفاژی نرمال داشته باشد.

The effect of 4 weeks of continuous aerobic training with different volumes and starvation on the expression of some genes involved in liver mitophagy in healthy male Wistar rats

چکیده English

Introduction and Purpose: An increase in the number of defective mitochondria and their persistence can lead to apoptosis and necrosis of cells. Regular physical exercise along with time-restricted eating (TRE) can play an important role in improving mitochondrial damage and function. Therefore, the aim of this study was to investigate the effects of 4 weeks of continuous aerobic exercise and fasting on the expression of some genes involved in mitophagy, including PINK1 and BNIP3, and some factors influencing liver health such as liver enzymes ALT and AST, and the lipid profile in healthy rats.
Materials and methods: his was an experimental study. Thirty healthy male Wistar rats, 18–20 weeks old, with an average body weight of 330.40 ± 25.34 grams, purchased from the Pasteur Institute of Iran, were randomly assigned to six groups of five: Control, Fasting, 3 days of exercise, 5 days of exercise, Fasting + 3 days of exercise, Fasting + 5 days of exercise. The exercise program was performed on a treadmill for 4 weeks, 3 or 5 days per week, for 45–60 minutes per session. Tissue sampling was conducted 24 hours after the last exercise session following a 14-hour fasting period, and blood samples were collected directly from the heart. One-way ANOVA and LSD post-hoc tests were used for data analysis.
Results: Weight, cholesterol, and ALT levels significantly decreased in all experimental groups compared to the control group (P≤0.05). The groups "fasting + 3 days of exercise" and "fasting + 5 days of exercise" showed greater reductions in these indices compared to other groups, but there was no significant difference between the two. Triglyceride levels also significantly decreased in all groups compared to the control group, with no significant differences observed among the other groups. AST levels significantly decreased in all groups compared to the control group (P≤0.05), with greater reductions observed in the "3 days of exercise," "5 days of exercise," "fasting + 3 days of exercise," and "fasting + 5 days of exercise" groups. Again, no significant difference was found among these groups.
The PINK1 marker significantly decreased in the "5 days of exercise," "fasting + 3 days," and "fasting + 5 days of exercise" groups compared to the control and fasting groups (P≤0.05), with no significant difference among the remaining groups. The BNIP3 marker showed a significant increase in the "3 days of exercise," "5 days of exercise," and "fasting + 3 days of exercise" groups compared to the control group (P≤0.05). The "5 days of exercise" group had a greater increase compared to the "fasting + 3 days of exercise" group. No significant differences were observed between the fasting and fasting + 5 days of exercise groups compared to the control.
Discussion and Conclusion: Four weeks of continuous aerobic exercise combined with fasting can positively impact liver health under normal mitophagy conditions by lowering lipid profile markers and increasing BNIP3 gene expression.
