تأثیر تمرین هوازی بر بیان ژن آسپروزین بافت پانکراس رت‌های نر مبتلا به دیابت

چکیده

مقدمه و هدف: آسپروزین یک هورمون پروتئینی ضدتنظیمی انسولین است و با بیماری‌های مزمن همچون چاقی و دیابت مرتبط است. از این رو هدف از پژوهش حاضر بررسی تأثیر تمرین هوازی بر بیان ژن آسپروزین بافت پانکراس رت‌های نر مبتلا به دیابت است.
مواد و روش‌ها: 32 سر رت نر ویستار، پس از القا دیابت با STZ به‌صورت تصادفی به 4 گروه کنترل (n=8)، تمرین (n=8)، دیابت (n=8) و دیابت + تمرین (n=8) تقسیم شدند. رت‌های گروه تمرین پس از آشنایی با پروتکل تمرین، 5 جلسه در هفته، به مدت هشت هفته، پروتکل تمرین هوازی را اجرا کردند. پس از اتمام پروتکل، بافت پانکراس استخراج و از روش RT- PCR برای اندازه‌گیری RNA و از روش الایزا برای سنجش بیان پروتئین آسپروزین استفاده شد. از آزمون تحلیل واریانس جهت تجزیه‌وتحلیل داده‌ها، در سطح معناداری (0.05>P < span lang="FA">) استفاده شد.
یافته‌ها: نتایج نشان داد القاء دیابت سطح بیان ژن و پروتئین آسپروزین در بافت پانکراس را به‌طور معنی­داری افزایش داد (0.001>P < span lang="FA">). هشت هفته تمرین هوازی بیان پروتئین آسپروزین را در گروه تمرین به‌طور معنی­داری کاهش داد (0.019=P < span lang="FA">) ولی کاهش در سطح بیان ژن و پروتئین آسپروزین در گروه دیابت +ت مرین نسبت به گروه دیابت معنی­دار نبود.
بحث و نتیجه گیری: تمرین هوازی سطح آسپروزین بافت پانکراس را که در اثر دیابت افزایش ‌یافته بود کاهش داد ولی قادر نبود آن را به سطح اولیه خود برگرداند. به نظر می­ رسد مسیر تأثیرگذاری ورزش مستقل از مسیرهای سلولی و مولکولی در بافت پانکراس است و بیشتر از طریق مسیرهای پیام‌رسانی سلولی در عضله نقش ایفا می‌کند و در رابطه با آسپروزین در سلول‌های تخریب‌شده بافت پانکراس در اثر ابتلا به دیابت علاوه بر ورزش نیاز به روش‌های کمک درمانی دیگر نیز وجود دارد.

The effect of aerobic training on expression of asprosin in pancreas of male rat diabetic

Introduction and purpose: Asprosin is a counter-regulatory protein hormone of insulin and is associated with chronic diseases such as obesity and diabetes. Therefore, the aim of this study is to investigate the effect of aerobic exercise on the expression of the asprosin gene in the pancreatic tissue of male rats with diabetes.
Materials and methods: 32 male Wistar rats were randomly divided into 4 groups: control (n=8), exercise (n=8), diabetes (n=8), and diabetes+exercise (n=8) after inducing diabetes with STZ. After getting familiar with the training protocol, the rats in the training group performed the aerobic training protocol 5 times a week for eight weeks. After completing the protocol, pancreatic tissue was extracted and RT-PCR method was used to measure RNA and ELISA method was used to measure asprosin protein expression. Analysis of variance test was used to analyze the data (P<0.05).
Results: The results showed that induction of diabetes significantly increases the expression level of asprosin gene and protein in pancreatic tissue (P<0.001). Eight weeks of aerobic training significantly reduced asprosin protein expression in the exercise group (P=0.019) but the decrease in gene and protein expression of asprosin in the diabetes+exercise group was not significant compared to the diabetes group.
Discussion and Conclusion: Aerobic exercise reduced the level of asprosin in pancreatic tissue, which was increased due to diabetes, but was not able to return it to its original level. It seems that the effect of exercise is independent of the cellular and molecular pathways in the pancreatic tissue and plays a role mostly through the cell signaling pathways in the muscle, and in the damaged cells of the pancreatic tissue due to diabetes, in relation to asprosin addition to exercise, it is necessary to other treatment methods.
